What is nicotinamide mononucleotide?

Nicotinamide mononucleotide (NMN) is the molecule one step before NAD+. The body makes it from nicotinamide or NR, and it's studied, and sold, as a way to raise NAD+.

Definition

Nicotinamide mononucleotide, in plain English

NMN is a nucleotide made of nicotinamide, a ribose sugar and a phosphate. In the salvage pathway, the enzyme NAMPT makes NMN from nicotinamide, and NMNAT enzymes then convert NMN to NAD+.

NMN is sold mainly as an oral product. In 2022 the FDA said NMN couldn't be sold as a dietary supplement; in September 2025 it reversed that view after a citizen petition.

How it connects

A 2021 trial in women with prediabetes found that 10 weeks of NMN increased muscle insulin sensitivity by about 25%, according to an NIH summary of the study (Yoshino et al., Science).

What nicotinamide mononucleotide means for NAD+ therapy

NMN is mostly taken by mouth, while NADMedics prescribes NAD+ itself. Your provider can talk through how the options differ; none is proven to slow aging.

Questions

Nicotinamide mononucleotide: common questions

What is nicotinamide mononucleotide?

Nicotinamide mononucleotide (NMN) is the molecule one step before NAD+. The body makes it from nicotinamide or NR, and it's studied, and sold, as a way to raise NAD+.

Is NMN the same as NAD+?

No. NMN is the direct precursor; one enzyme step turns it into NAD+.

Does NMN work?

Small human trials show it can raise NAD+ markers, and one found better muscle insulin sensitivity in women with prediabetes. There's no proof it slows aging.

Can I take precursors with NAD+ therapy?

Combining them hasn't been studied. Tell your provider about any NR, NMN, niacin or other supplements before you start.

Can I get enough NAD+ from food?

Food supplies niacin and tryptophan, which your body turns into NAD+, and NIH says most people in the U.S. get enough niacin. NAD+ in food is largely broken down during digestion.

Do precursors and NAD+ therapy do the same thing?

Both aim to raise NAD+, by different routes: precursors are converted by the body, while spray and injections deliver NAD+ itself. They haven't been compared in trials.
